Like it says in the Current User Settings tool "Run at start up", meaning, it runs when you start your computer. If you uncheck it, it just prevents it from running at startup, it doesn't close the program.
Also, you could use the uninstaller to uninstall it from your system, or, press ctrl+alt+del to open task manager and seen it listed right there as "Alt+Q Hotkey.exe", and just ended the process.
